Determination of Genetic Stability in Cacao Plants (<i>Theobroma Cacao</i> L.) Derived from Somatic Embryogenesis Using Microsatellite Molecular Markers (SSR)

The clonal propagation of T. cacao by somatic embryogenesis (SE) is a promising approach to multiply elite genotypes. Assessing fidelity in plants regenerated from embryos the first step toward ensuring genetic uniformity mass production planting material. This study assessed stability plantlets propagated SE and conventional grafting for genotypes CCN51 TSH565 using 13 SSR. leaves vitro (IVL) were collected 6-month-old field (FPL) selected 3-year-old trees. analyzed loci revealed 25 alleles genotype 24 TSH565. highest PIC value was observed all SSR, only mTcCIR8 mTcUNICAMP09 intermediate, with values less than 0.250. IVL FPL populations genetically equal. According results, no differences allelic composition between each genotype. indicates that did not show perceptible detriment their genome used In addition, Jaccard’s coefficient showed more 91% similarity 92% CCN51. UPGMA PCA tended group within two SSR results obtained do exclude occurrence other changes nuclear genome. However, considering morphological plants, indicate protocol suitable efficient large scale, true-to-type commercial purposes.
